Study the Biochemical active materials (Allicin, Alliin) extraction from medical plant to treat patients with atherosclerosis

otal study was concerned with the Iraqi garlic extract which should concentration of both Allicin 23.94 ppm and Alliin 17.9 ppm respectively. The highest concentration of Arginine from sixteen amino acid and is the highest concentration of vitamin (Niacin) B3 in garlic extract. The effect of some Biochemical variables for patients atherosclerosis was studied at the age between (45-65) years old using the 3 capsules a day to give a concentration 250mg/ capsule for a period of two weeks each at the level of total cholesterol, HDLc, and MDA and the effectiveness of the enzyme G.S.T. The results indicate a low level of total cholesterol, MDA and the level of effectiveness of the enzyme G.S.T. a rise in the level of HDLc.

تم دراسة المجاميع الفعالة لمستخلص الثوم العراقي و كان مستوى كل من الاليسين ppm 23.94 و الاليين ppm 17.9 . وكان أعلى تركيز لحامض الارجينين من ستة عشر حامض أميني موجود و أعلى تركيز لفيتامين (Niacin) B3 في مستخلص الثوم . تم دراسة تأثير بعض المتغيرات البايوكيميائية لمرضى تصلب الشرايين من خلال إعطاءهم 3 كبسولات يوميا بتركيز 250 ملغم / كبسولة لمدة اسبوعين وكان عمر المصابين يتراوح من (45-65) سنة على مستوى كل من الكولسترول الكلي ، HDLc و MDA وفعالية أنزيم G. S. T . اشارت النتائج بانخفاض مستوى الكولسترول الكلي و MDA ومستوى فعالية أنزيم G.S.T وارتفاع في مستوي HDLc .

The objective of the experiment, which conducted under laboratory conditions was to investigate the interaction effects of using three levels of Molybdenum (0, 0.25, 2.50) mg Mo .L-1 and four levels of potassium (0, 25, 50, 100) mg K.L-1 on the optical density of Rhizobium growth in the liquid medium (broth media), which is belong to R. leguminosarum after three days of incubation at 28°C.

liadin protein of three soft wheat cultivars namely Abu-Ghraib, IPA 99 and AL-Littefia was extracted and separation by high performance liquid chromatography HPLC. Result showed that there were significant differences between the gliadin percentage ratio with the order of AL-Littefia 34.02% , IPA99 30.20% and Abu-Ghraib 31.00% . Also the result of HPLC analysis revealed that the gliadin of the test cultivars has 3 distinguish patterns of separation. Thus it appears that HPLC provides efficient tool for identification of wheat cultivars. Abu-Ghraib gliadin showed 6 peaks while IPA99 and AL-Littefia showed 15 and10 peaks respectively

تم استخلاص وفصل بروتين الكلايدين لثلاثة أصناف من الحنطة الناعمة المعتمدة وهي أبو غريب وإباء 99 واللطيفية باستخدام الفصل الكروموتوكرافي High performance liquid chromatography هدف تقدير كميته وتوضيح إمكانية استخدام هذه التقنية كوسيلة تعريف مثبتة لهوية هذه الأصناف واستخدامه كطبع الإصبع fingerprint في التمييز بين هذه الأصناف . وبينت النتائج وجود فروق معنوية بين قيم النسب المئوية للكلايدين من البروتين الكلي وكان طحين حنطة اللطيفية هو الأعلى في محتواه من الكلايدين من البروتين الكلي إذ بلغ 34.02 % مقارنةً بصنف إباء 99 الذي كان 30.20 % وصنف أبو غريب 31.00 %. ووضحت نتائج الـ HPLC إن هناك ثلاثة أنماط مختلفة لكلايدين الأصناف الثلاثة إذ اظهر كلايدين صنف أبو غريب 6 قمم واضحة بينما كان لصنف إباء 99 15 قمة ولصنف اللطيفية 10 قمم

total of 62 different Streptomyces isolates were recovered from 17 samples of soil collected from different sites of Nineveh Province. Only 23 isolates showed activity against test Gram-positive and Gram-negative bacteria. One isolate showed high antimicrobial activity against Staphylococcus aureus, was selected and identified as Streptomyces lavendulae on the bases of microscopic, morphologic, biochemical tests and its sensitivity to some antibiotics. The bioactive compound produced by S. lavendulae was isolated on TLC plate (Rf 0.85). The UV spectrum of the active compound in methanol showed one peak at 280 nm. From these data it could be concluded that the active compound probably belongs to macrolide antibiotics group.

تم الحصول على 62 عزلة مختلفة لجنس Streptomyces من 17 عينة ترب جمعت من مواقع مختلفة لمحافظة نينوى بينت 23 عزلة فقط نشاط ضد البكتيريا المختبرة من الأنواع الموجبة و السالبة لصبغة كرام . ابدت عزلة واحدة نشاط تضاد عالي ضد البكتيريا Staphylococcus aureus . تم اختبار هذه البكتيريا و شخصت بكونها Streptomyces lavendulae على اساس الأختبارات الميكروسكوبية والشكلية و الكيموحيوية وحساسيتها لبعض المضادات الحيوية . تم عزل المركب ذو النشاط الحيوي الذي تنتجه عزلات S. lavendulae على لوح كروماتوغرافيا الطبقة الرقيقة ، وكان Rf للمركب 0.85 . بين طيف الأشعة فوق البنفسجية للمركب الفعال المذاب في الميثانول ذروة واحدة فقط عند 280 نانوميتر . وبالأعتماد على هذه البيانات يمكن الأستنتاج ان المركب الفعال من المحتمل ان يعود الى مجموعة الماكروليد من المضادات الحيوية

he effect of licorice extract [Glycyrrhiza glabra] on somatic cells chromosomes from 30 Awassi ewes (local breed) before and after pregnancy to improve soil fertility through the use of chromosome analysis of lymph blood cells and on their embryos from amniotic cells chromosome which drain by laparotomy of the maternal and fetal. That is the aim of this study. The blood taken from ewes treatment [0.5ml] for each sample incubate in media Tc-199 with Brdu for 72 hours worked in Cytogenetic analysis of the models Rd cells of embryos derived from the total ewes experience a way to open the abdomen of the pregnant ewes )12-13( weeks to demonstrate the effects of licorice extract chromosomes in blood lymphatic cells of mothers and fetuses after treatment. Cytogenetic analysis revealed that licorice extract could not induce numerical nor structural abnormalities in chromosomes from ewes blood lymphocytes, after treatment or on their embryos from amniotic cells chromosomes which drain by laparotomy under their experimental designs.

أجريت التحليلات الخلوية الوراثية لنماذج الدم المأخوذه من نعاج عواسية( محلية ) 0.5 مل لكل نموذج ، حضنت في وسط زرعي TC-199 مع Brdu لمدة 72 ساعة فيما عملت التحليلات الخلوية الوراثية لنماذج خلايا سائل السلي للاجنة المأخوذه من مجاميع نعاج التجربة(n=30) بطريقة فتح البطن للنعاج الحوامل ( 12 – 13 ) اسبوع . كان هدف الدراسة لبيان التأثيرات السمية لمستخلص عرق السوس في كروموسومات خلايا الدم اللمفاوية للامهات والاجنة بعد المعاملة . بينت التحليلات الخلوية الوراثية عدم احداث تغير عددي او تركيبي كروموسومي للامهات واجنتها المعاملة بمستخلص عرق السوس وتحت ظروف التجربة .

he study was carried out in order to determine the toxic, oxidant and antioxidant effects for one compounds of the Curcrcuma longa L. in comparison with vitamin C against the mutagenic effect of carbon tetrachloride CCl4, which is a chemical compound that damages hepatic cells and has mutagenic effects. The effect was studied in a mammalian system. This in vivo system depended on evaluating the enzymatic activity of two anti oxidant enzymes: Catalase and Glutathione reductase. Two concentrations of pure curcumin 200 and 400 mg /kg were evaluated to chose the suitable concentration, which remembered the negative control. In order be used in The interaction experiments, included two types of treatments pre – CCl4 and post-CCl4 in order to determine the mechanisms of the pure curcumin in preventing or reducing the antioxidant effect of CCl4. The pure curcumin showed no toxic and oxidant effects in biological system, and instead it showed highly performance in preventing or reducing the oxidant stress effects of CCl4. It increased the Catalase and Glutathione reductase antioxidant activity, especially in dose 200 mg/kg. The positive effect was higher when pure curcumin was used as pre-CCl4 treatments, and to less extent in post-CCl4 treatment. Therefore, the plant extracts can be considered as adesmutagen at the first degree and a bioantimutagen at the second degree.

أجريت الدراسة للكشف عن ألتأثير السمي والمؤكسد والمضاد للأكسدة لاحد مركبات نبات الكركم Curcrcuma longa مركب الكركمين النقي ومقارنته بفيتامينC تجاه رباعي كلوريد الكاربون CCl4 , والذي يعد مركب كيميائي يسبب تلف خلايا الكبد ويمتلك تأثيرا تطفيريا , وباستخدام نظام اللبائن في in vivo وبالاعتماد على تقييم فعالية الانزيمية لمضادين للأكسدة: الكاتاليز و كلوتاثايون المختزل لمجانس خلايا الكبد . استخدم تركيزين للكركمين النقي (200 ،400) ملغرام/كيلوغرام وكل على انفراد لاختبار فعلها المؤكسد والمضاد للأكسدة في الفئران البيض . وتم انتخاب التركيز الأمثل للمركب والذي أعطى نتائج أفضل من الحالة الطبيعية السيطرة السالبة ، بعد ذلك اجري التداخل ما بين التركيز الأمثل والمطفرCCl4 وبشكل معاملتان قبل وبعد المطفر لمعرفة الآلية التي يعمل بها هذا المركب في منع أو تقليل الأثر ألتأكسدي للمطفر ، توصلت الدراسة إلى عدم إظهار الكركمين النقي تأثيرات سمية و تأكسدية في التركيزين المستعملين وعلى مستوى النظام الحيوي المستخدم , واظهر الكركمين النقي كفاءة عالية في تقليل الأثر التأكسدي للمطفر CCl4 فقد عمل على رفع قيمة إنزيمي الكاتاليز الكلوتاثايون المختزل المضادين للأكسدة , وقد كان الفعل الأكثر ايجابية عند استعمال الكركمين النقي بجرعة 200 ملغم/ كغم قبل المطفر وبدرجة اقل عند معاملة الحيوانات بالكركمين النقي بعد المطفر وبالتالي يمكن تصنيف فعل هذا المركب في نظام اللبائن كونه مثبط مباشر Desmutagens بالدرجة الأولى ومثبطات حيوية Bioantimutagens بالدرجة الثانية .

study was conducted at the tissue culture lab. College of Agriculture/ University of Baghdad and Drug Research Center/ Health Minstry during 2006 - 2007. Results indicated that MS medium supplemented with 30 g/l of sucrose gave the highest values (630.3, 61.6) mg of both fresh and dry weight of callus respectively. However, values of both parameters were reduced with increasing sucrose concentrations up to 60, 90, 120 g/l (478.2, 346.5, 217.6) mg respectively. MS medium supplemented with 30 mg/l of tyrosine gave the highest values of callus fresh and dry weight (429.7, 42.6) mg respectively. MS medium supplemented with 30 g/l of sucrose and 30 mg/L tyrosine gave the highest values (688.0, 67.5) mg of both fresh and dry weight callus. MS medium supplemented with 30 mg/l of tyrosine and 90 g/l of sucrose gave the highest values (2.9, 2.8) mg/g of morphine and codeine from fresh callus weight respectively while the lowest values of morphine and codeine (0.5K 0.2) mg/g from fresh callus weight respectively of control treatment .

اجريت الدراسة في مختبرات زراعة الانسجة النباتية – كلية الزراعة – جامعة بغداد ومختبرات مركز الرقابة والبحوث الدوائية في وزارة الصحة خلال عامي 2006-2007 . اعطى الوسط MS المجهز تركيز 30 غم/لتر سكروز اعلى معدل للوزن الطري والجاف للكالس بلغ (630.0 ، 61.6) ملغم على التوالي . وانخفض معدل الوزن الطري والجاف للكالس بزيادة تركيز السكروز الى (60 ، 90 ، 120) غم/لتر فبلغ (478.2 ، 346.5 ، 217.6) ملغم للوزن الطري و (46.8 ، 34.3 ، 21.0) ملغم للوزن الجاف على التوالي . وتشير النتائج الى ان وسط MS المجهز بتركيز 30 ملغم/لتر تايروسين اعطى اعلى معدل وزن طري وجاف للكالس بلغ (429.7 ، 42 ، 6) ملغم على التوالي . وبينت النتائج ان الوسط MS المجهز بتركيز 30غم/ لتر من السكروز و 30 ملغم/لتر تايروسين قد اعطى اعلى معدل للكالس على اساس الوزنين الطري والجاف بلغ (688.0 ، 67.5) ملغم على التوالي . وتفوق الوسط MS المجهز بتركيز 90 غم/لتر سكروز وسجل اعلى قيمة عند التركيز 30 ملغم/لتر من التايروسين لقلويدي المورفين والكودائين فبلغ (2.9 ، 2.8) ملغم/غم وزن طري للكالس على التوالي . بينما كانت اقل القيم للمورفين والكودائين عند معاملة المحايد بلغ (0.5 ، 0.2) ملغم/غم من الكالس الطري على التوالي .

he effect of some transition metals, Nickel, Copper, Zinc and Selenium elements on Eruca sativa seeds germination was studied. Concentration (1, 10, 100, 500, 1000) ppm in sterile distilled water were used to cover the natural concentrations and the concentrations in polluted environments. Germination was recorded after 5 days. Results showed that there were inverse relationships between the number of germinated seeds and increasing in concentration of elements. The effect was mainly due to the elements, while the pH effect was very low and not exceeds 7 % except for Zinc solution which was about 50%. The correlation coefficient between germination and concentration were with negative values, it was for Nickel element -0.842, Copper -0.965, Zinc -0.928 and for Selenium -0.603. The values were statically significant except for selenium. According to the obtained results the elements can be ranked Nickel the less effect < Copper < Zinc < Selenium.

تمت دراسة تأثير بعض العناصر الانتقالية وهي النيكل ، النحاس ، الزنك وعنصر السيلينوم في انبات بذور نبات الجرجير Eruca sativa في الماء المقطر المعقم بتراكيز متدرجة ( 1 ، 10 ، 100 ، 500 ، 1000) جزء بالمليون لتغطي التراكيز في البيئات الطبيعية وكذلك التراكيز في البيئات الملوثة ، استمرت عملية التنبيت لمدة 5 أيام . ثم سجل عدد البذور النابتة . أشارت النتائج إلى تأثير عملية الانبات بشكل سلبي عند زيادة التراكيز . وكان التأثير الاكبر للايونات اما التغير في الارقام الهيدروجينية فكان قليلا" ولم يتجاوز 7 % الا في حالة الزنك اذ بلغ 50 % . كان معامل الارتباط بين مؤشر الانبات والتركيز سالبا" على مدى التراكيز المستعملة ، اذ بلغ بالنسبة للنيكل -0.842 ، النحاس -0.965 ، الزنك -0.928 والسيلينوم -0.603 ولذا يلاحظ ان العلاقة كانت معنوية ( P ≤ 0.05) ما عدا في حالة السيلينوم . ومن النتائج التي تم الحصول عليها يمكن تدريج العناصر من الاكثر تأثيرا ( سلبي ) الى الاقل بالاتي : النيكل > النحاس > الزنك > السيلينوم

تمت دراسة تأثير الليزر في بعض جوانب الأداء الحياتي لحشرة عثة التين المشععة و يرقات بعمر 5-6 أيام و عذارى بعمر يوم واحد و خمسة أيام وأوضحت النتائج أن لأشعة الليزر تأثير معنوي في مدة الدورين اليرقي والعذري ونسبة خروج البالغات ومدة حياتها لوحظ أنه كلما تقدمت اليرقات والعذارى المعرضة لأشعة الليزر بالعمر فأن التأثير يكون أقل ، فضلاً عن ذلك أشارت النتائج تأثر قابلية وضع البيض ونسبة فقسه في التزاوجات المختلفة التي أجريت وكلما ازدادت الجرعة الإشعاعية .

Preparation of shrimp shells chitosan and use it in extending eggs shelf life

his study aimed to examine the performance of chitosan prepared from chitin. By treating the shrimp shells with 3.5% sodium hydroxide then with 1 N hydrochloric acid. The chitin produced was used to prepare two types of chitosan by treating it with concentrated solution of sodium hydroxide 50% with heating at 100c°for 4 hr. (type A) and 20 hr. (type B). The molecular weights of samples A and B as determined by viscometer were 1155 and 720 KDa. The performance of the two chitosans was examined in preserving eggs quality and extending its shelf life after coating them with 2%of chitosan and keep it at 25C°for 4 week. The results indicate that both samples showed better performance than control (non-coated eggs and eggs coated with 1% acetic acid) as A and B contributed in preserving albumen and yolk quality, whereas, Hough Unit of coating sample decreased from 100 (in the 1st day) to (76.3, 76.8), respectively after 4 wk of storage while the control (non coated eggs and 1% acetic acid coated eggs) was (68.2, 67.9), respectively. The yolk index decreased from 0.6 (in 1st day) to 0.27 and 0.28 for eggs coated with chitosan A and B, respectively and to 0.21 and 0.22 for none coated and 1% acetic acid coated eggs respectively after 4 week of storage. A and B coated samples contributed in decreasing the albumen pH increasing which increased from 7.9 ( in the first day) to 10.4 and 10.8, respectively as compared to 11 for the control. B sample was the best compared to A sample and control in preventing decreasing weight losses which were (5.51, 6.12, 6.33, 7.69) %, respectively. Therefore A and B coated eggs showed high activity to improve the internal quality of eggs and to extend its storage period

هدفت الدراسة الحالية إلى اختبار أداء الكايتوسان المحضر من مخلفات الروبيان في اطالة العمر الخزني للبيض المحلي . إذ حضر الكايتين بمعاملة قشور الروبيان بمحلول هيدروكسيد الصوديوم 3.5% ثم بمحلول حامض الهيدروكلوريك 1عياري واستخدم الكايتين الناتج من تحضير نوعين من الكايتوسان ، من خلال معاملة الكايتين بمحلول مركز من هيدروكسيد الصوديوم 50% والتسخين بدرجة حرارة 100م لفترات زمنية (4 ،20) ساعة رمز لهما A و B على الترتيب . قدر الوزن الجزيئي بطريقة مقياس اللزوجة للنموذجين A و B فكانا (1155 , 720) كيلودالتون . تم اختبار أداء النموذجين بتركيز 2% في المحافظة على نوعية البيض وإطالة العمر ألخزني له وذلك بتغليف البيض بهما ثم حفظها بدرجة حرارة 25م لمدة 4 أسابيع . أشارت النتائج المستحصلة إلى إن النموذجين اظهرا أداء أفضل في إطالة العمر ألخزني للبيض المحلي بالمقارنة مع نموذجي القياس ( البيض غير المغلف ، والمغلف بمحلول حامض ألخليك1% ) إذ ساهم كل من A و B في الحفاظ على نوعية الألبومين والصفار للبيض المغطى بهما ، حيث انخفضت قيم وحدة ألهو Haugh unit للعينات المغطاة بهما من 100 (لليوم الأول ) إلى (76.3 ، 76.8) على الترتيب بعد 4 أسابيع من الخزن في حين كانت لنموذجي القياس ( البيض غير المغلف والمغلف بمحلول حامض ألخليك 1%) ( 68.2 ، 67.9) على الترتيب بعد 4 أسابيع من الخزن . إما معامل الصفار Yolk index فقد انخفض من 0.6) لليوم الأول) إلى ( 0.27 ، 0.28) للعينات المغطاة بالكايتوسان A و B والى القيمة (0.22 ، 0.21 ) لنموذجي القياس بعد 4 أسابيع من الخزن . ساهمت النماذج A و B في خفض الارتفاع الحاصل في الرقم الهيدروجيني لألبومين البيض المغلف بهما إذ ازداد الرقم الهيدروجيني من 7.9 (لليوم الأول) إلى ( 10.4 ، 10.8 ) على التوالي بعد أربع أسابيع من الخزن في حين بلغت في عينات نموذجي القياس 11 . تفوق نموذج B على نموذج A ونموذجي بيض القياس في خفض مقدار الفقد بالوزن حيث بلغ ( 5.51 ، 6.12 ،6.33 ، 7.69) % على التوالي بعد أربعة أسابيع من الخزن . وبذلك أظهرت أغطية الكايتوسان A و B فعالية عالية في تحسين النوعية الداخلية للبيض وإطالة مدة حفظه

Partial Purification and Characterization of Inulinase Produced from a local isolate of Aspergillus niger J3

nulinase was produced from local isolate of Aspergillus niger J3. The inulinase was purified by two steps included precipitation by amonium sulphate at (30-80) % sat-uration and gel filtration on sephadex G100. The final purification folds and the yield of the enzyme were 3.15 times and 28.24%, respectively. The purified enzyme has the following characteristics: The optimum pH of the enzyme activity was 5.5. The enzyme was most stable at pH (4.5 - 6). The optimum temperature for its activity was 45c. The enzyme retained its original activity when incubates at (30-55) c for 20 minutes. Mercury chloride inhibited the enzyme completely at concentration of 10mM, cupper sulphate and calisium chloride inhibited the enzyme at concentrations of 85% and 7% respectively. It was revealed that the enzyme had the efficiency to hydrolyze 87% of 5% inulin solution when treated at 45c for 120 min

انتج انزيم الانيولينيز من العزلة المحلية للعفن Aspergillus niger J3 وتم تنقيته بخطوتين شملت الترسيب بكبريتات الامونيوم بنسبة اشباع (30 -80)% ومن ثم الترشيح الهلامي في sephadex G100 وبلغت عدد مرات التنقية بعد هذة الخطوات 3.75 مرة وبحصيلة انزيمية مقدارها 33.65% . وقد بينت نتائج توصيف الانزيم بان الرقم الهيدروجيني الامثل لفعالية الانزيم 5.5 وتراوح الرقم الهيدروجيني الامثل لثبات الانزيم بين ( 4.5-6) ووجد ان درجة الحرارة المثلى للفعالية تبلغ 45م واحتفظ الانزيم بكامل فعاليته عند حضنه مدة 20 دقيقة في مدى من الدرجات الحرارية تراوح (30 -55)م . ثبط كلوريد الزئبق فعالية الانزيم بالكامل عند التركيز 10 ملي مولاري في حين ثبطت كبريتات النحاس و كلوريد الكالسيوم فعالية الانزيم بنسبة ( 85 , 7)% على التوالي عند نفس التركيز . تميز مستخلص الانزيم بكفاءة لتحليل محلول الانيولين بتركيز 5% حيث بلغت نسبة التحلل 87% بعد 120 دقيقة من المعاملة الانزيمية وبحرارة 45 م

his present study was conducted to evaluated the antibacterial activity of crude extracts (Methanolic) of punica granatum peel, Menthe longifolia and Olea eurpeae leaves against some gram negative bacteria like Pseudomonas aerugenosa, Serratia marcesens, Escherichia coli, Proteus vulgaris, Klebsiella pnemunoniae. The Agar-well diffusion was used to know the ability of inhibititory of crude extracts against bacteria. Results showed different susceptibility response of bacteria under study. It was observed that Pseudomonas aerugenosa the most susceptible to Methanolic extracts to all plants in (25, 50) mg/ml-1 concentrate. While the bacterial strains shown no responding against mint and pomegranate extracts in 10 mg/ml-1 except Pseudomonas aerugenosa and Escherichia coli, was inhibited with Olive extracts only. That ability to inhabitation less in concentrate 10 mg ml-1.

أجريت هذه الدراسة لتقدير التاثيرالتثبطي لمستخلصات الخام ( الميثانولية) لقشور الرمان ، Punica granatum و أوراق ا لنعناعMentha longifolia والزيتونOlea eurpeae على عزلات مختلفة من البكتريا الممرضة السالبة لصبغة كرام متمثلة بــcoli Pseudomonas aerugenosa, Serratia marcesens Escherichia Proteus vulgaris, Klebsiella pnemunoniae. إذ استخدمت طريقة الانتشار بالحفر لمعرفة القدرة التثبيطية للمستخلصات أعلاه تجاه النمو البكتيري . أظهرت النتائج وجود تباين في تحسس الأنواع البكتيرية اذ لوحظ أن بكتريا Pseudomonas aerugenosa أكثر تحسسا ًللمستخلصات الميثانولية لجميع النباتات عند تركيز (25,50) ملغم / مليلتر . بينما لم يبين أي من الأنواع البكتيرية استجابة لمستخلصي النعناع والرمان عند تركيز 10ملغم/مليلتر باستثناء بكتريا E.coli , Pseudomonas aerugenosa التي ثبط نموهما فقط بمستخلص الزيتون كانت القابلية التثبيطية له أقل عند تركيز 10ملغم / مليلتر .

The aim of this research is to study the effects of salvia aqueous extract on some parameters of sperms and histopathological changes in testis of mice treated with methotrexate. Twenty-four adult albino male mice were divided into three groups, (8 mice in each group) First group was negative control received normal saline intraperitoneally, while second group was treated with50 μg/kg, for 35 days intrapritoneally (ip). The third group was treated with methotrexate (50 μg/kg) also (ip) and extract of salvia 85 mg/kg for 35 days. The results showed significant (P<0.05) decrease in sperms motility , increased percentage of dead sperms and abnormalities of sperms in mice treated with MTX, there was significant (P<0.05) decrease in diameters of seminiferous tubules, primary spermatocytes and spermatids, increased interstitial spaces as compared with control, while in the third group result showed significant (P<0.05) increase in sperms motility , decreased percentage of dead and abnormalities of sperms and significant (P<0.05) increase in diameters of seminiferous tubules, primary spermatocytes and spermatids and decrease interstitial spaces compared with first and second groups.

ان الاستخدام الشائع لعقار الميثوتركسيت هو لعلاج الامراض السرطانية ، الهدف من هذا البحث هو دراسة تاثير المستخلص المائي للمريمية على بعض قياسات الحيامن والتغيرات النسيجية المرضية لخصى الفئران المعاملة بالميثوتركسيت ، 24 ذكر ناضج من الفئران البيض قسمت الى ثلاث مجاميع كل مجموعة تحتوي على 8 ذكور . المجموعة الاولى السيطرة السالبة تم اعطائها في الخلب البريتوني المحلول الملحي الفسيولوجي ، بينما المجموعة الثانية عوملت بعقار الميثوتركسيت بجرعة 50 مايكروغرام/كغم لمدة 35 يوما عن طريق الخلب البريتوني . المجموعة الثالثة فعوملت بعقار الميثوتركسيت بجرعة 50 مايكروغرام / كغم عن طريق الخلب البريتوني وعولجت بالمستخلص المائي لنبات المريميا لمدة 35 يوما. اظهرت النتائج انخفاضا معنويا في حيوية الحيامن ، وزيادة معنوية في نسب الحيامن الميتة والمشوهه وكذلك انخفاض معنوي في اقطار النبيبات المنوية ، وخلايا النطاف الاولية والنطاف ، وزيادة في المسافات البينية للنبيبات المنوية لخصى الذكور المعاملة بالميثوتركسيت مقارنة مع مجموعة السيطرة السالبة ، بينما اظهرت النتائج زيادة معنوية في حيوية الحيامن ، وانخفاضا معنويا في نسب الحيامن الميتة والمشوهه وكذلك انخفاضا معنويا في اقطار النبيبات المنوية ، وخلايا النطاف الاولية والنطاف ، ونقصان في المسافات البينية للنبيبات المنوية لخصى الذكور في الفئران المعالجة بالمستخلص المائي للمريميا مقارنة مع المجموعة الاولى والثانية .

he aim of this research is to establish a gene mutation assay for examining the integrity of animal cell genome for nuclear transfer technique. Lung fibroblasts which were expanded from 4 months old female lamb were selected as target cells. These cells were coded (SLF) as Sheep lung cells. Growth characterization, doubling time, chromosomal number and structural integrity were checked after their growth in RPMI-1640 medium. For HPRT-gene mutation assay, the cells were plated at density of 1×103cells/plate and grown in medium containing toxic concentrations of 6-thioguanine; while for DHFR -gene mutation assay, toxic concentration of methotrexate was used as a selective agent. Those cells were grown for 15 days; mutant colonies either 6TGr or MTXr were reinoculated in a selective medium for further 8 weeks for checking the stability of phenotypic expression of mutant cells. The results revealed that SLF cells has spontaneous frequencies of HPRT, and DHFR gene mutations equal to 16 and 22×10-3 event/ generation/ cell, respectivel.These levels are normal comparing with other animal cell types, and these assays could be applied on other somatic cells.

كان الهدف من هذا البحث هو ايجاد طريقة Biomarker لفحص الطفرات الوراثية لجين الـHPRT والـ DHFR كمؤشرات حيوية تفيد في فحص سلامة الذخيرة الوراثية للخلايا الجسمية المراد استعمالها في عمليات نقل الانوية . استعملت خلايا رئة الاغنامsheep lung fibroblasts (SLF) التي سبق وان عزلت من رئة انثى حمل بعمر 4 اشهر ، وتمت تنميتها مختبرياً لمدة عام كامل في مختبرات منظمة الطاقة الذرية . درست خصائص النمو، زمن التضاعف والهيئة الكروموسومية التركيبية والعددية للخلايا بعد تنميتها في الوسط الزرعي (RPMI-1640) . ولغرض فحص التردد التلقائي للطفرة الوراثية لجين الـ HPRT ، تمت زراعة الخلايا بعدد 1000 خلية/طبق وفي وسط زرعي حاوي على التركيز القاتل من نظير الكوانين (6-Thioguanine 6TG) ولمدة 15يوماً . بينما تمت تنمية الخلايا للفترة ذاتها في وسط زرعي حاوي على التركيز القاتل من مادة الـMethotrexate (MTX) لغرض الكشف عن التردد التلقائي للطفرة لجين DHFR ولغرض التأكد من ظهور فعل الجين وبثبوتية الطفرة ، احتسبت المستعمرات الطافرة لكل جين وتمت اعادة زراعتها في الوسط الزرعي المنتخب لكل طفرة ولمدة 8اسابيع . تشير النتائج إلى ان خلايا رئة الاغنام المستعملة في هذا البحث تحوي على مستوى تلقائي من الطفرة في جين HPRT وDHFR تساوي 16 و22 ×10-3 حدث/جيل/الخلية . وهو مساوي إلى ذلك المكتشف في خلايا حيوان ألهامستر المستخدمة في المختبرات عالميا . يمكن تطبيق هذه التقنية على خلايا جسمية اخرى .

he present research concentrated on evaluating the biological activity of Colocynthis (Citrillus colocynthis) plant oil extract on peripheral blood cells from leukemic patients using some cytogenetic parameters. when it is added to this cell in culture media in different concentration and incubated for 72 hours at 37C, the results pointed out that cell exposed to the Citrillus extract in culture media have mitotic index near to normal ranges, and number of chromosomal aberrations in cells exposed to the Citrillus extract is less than in the leukemic cells. This study considered as an attempt in survey for anti-tumor agents in medicinal plants.

ixty five soil samples and fifteen water samples were collected from different places in which previously explosions were occurred in Iraq. Seven isolates showed ability to utilize 0.1mM trinitrotoluene (TNT) and/or 0.2mM glycerol trinitrate (GTN) as a sole carbon and nitrogen source and one of these isolates showed the highest nitrate reduction which was classified and coded as Pseudomonas sp. SH7. The highest nitrate reductase activity extracted by sonication while optimum conditions for enzyme production in minimal media pH 7 containing 0.25mM GTN at 35oC for 3 days under aerobic condition. Nitrate reductase was purified by 40-60% ammonium sulphate, ion exchange and gel filtration. Nitrate reductase molecular weight determined by SDS-PAGE was 115 kD. The characterization of purified enzyme activity and stability was higher at a pH between 6.5-7.5 and. Maximum activity was at 35oC and stable at 30-40oC for 15 min., while for heat sensitivity 100% activity observed at 45oC for 20 min. Treatment with 200 µM azide and 500 µM cyanide inhibited the activity by 76 and 91% respectively.

جمعت خمسه وستون عينة تربه وخمسة عشر عينة من الماء من مختلف المناطق التي تعرضت للانفجارات في العراق . لقد تميزت سبع عزلات بأعطائها نسبه اختزال عالية mM 0.1 من (TNT) و mM 0.2 من (GTN) كمصدر كاربوني ونتروجيني وحيدين . وبعد عدة خطوات من الغربله الاوليه انتخبت العزله SH7 كونها العزله الاعلى اختزالا للنترات مقارنة مع بقيه العزلات هي من نوع Pseudomonas. ان اعلى فعاليه لانزيمNAR تم الحصول عليها عند معامله الخلايا بالامواج فوق الصوتية . بينما الظروف المثلى للانتاج كانت بتنمية الخلايا في الوسط الادنى المحتوي على 0.25 mM من (GTN) عند رقم هيدروجيني 7.0 وحضن بدرجة حرارة35 م لمدة ثلاثة ايام تحت ظروف هوائية . تضمنت خطوات تنقية ألــNAR الترسيب بكبريتات الامونيوم بنسبة اشباع 60-40% والتبادل الايوني والترشيح الهلامي . كما ان الوزن الجزيئي للانزيم 115 كيلو دالتون عند تعينه بأجراء الترحيل الكهربائي في هلام متعدد الاكريل امايد بوجود المادة الماسخة للبروتين SDS . واظهرت النتائج ان اعلى فعالية وثبات للانزيم كانت من الرقم الهيدروجيني 7.5-6.5. ولوحظ ان اقصى فعالية للانزيم كانت عند درجة حرارة35 م واحتفظ الانزيم بفعاليته عند حضن لمدة 15 دقيقة بدرجات حرارة40 – 30) )م . كما احتفظ الانزيم بكامل فعاليته عند حضن لمدة 20 دقيقة بدرجة حرارة45 oم. وقد وجد ان فعالية الانزيم قد ثبطت عند معاملته بــ200 µM من الازايد وµM 500 من السيانيد الى انخفاض في الفعالية بلغ 76)، (91% ، على التوالي .

This study involves the chemical synthesis of the novel ligand 5-(2-diphenylphosphino) phenyl-1,2-dihydro-1,2,4-triazole-3-thione (DPDTT) by the reaction of 2-diphenylphosphino benzoic acid with absolute ethanol that yield ethyl 2-diphenylphosphino benzoate and by cyclization of this compound with thiosemicarbazide, DPDTT will be produced. The chelating complexes of this ligand with Cr(III), Co(II), Ni(II), Cu(II) and Cd(II) were also prepared and studied. The new complexes were characterized by FT-IR, UV/visible spectra, and room temperature magnetic susceptibility. The stability for the prepared complexes was also measured using the density function theory and it was found that the cadmium complex is the most stable and the chromium complex is the least stable. Free ligand and its metal complexes have been tested in vitro against a number of microorganisms, like gram positive bacteria Staphylococcus aureus and gram negative bacteria E. coli, Proteus vulgaris, Pseudomonas and Klebsiella in order to assess their antimicrobial properties. All complexes showed considerable activity against all the studied bacteria.

تم تحضير معقدات جديدة للمشتق 5-(2-ثنائي فنيل فوسفينو) فنيل-12,-ثنائي هيدرو-4,2,1-ترايازول-3-ثايون (DPDTT)مع ايونات الفلزات: الكروم الثلاثي ، الكوبالت ، النيكل ، النحاس ، والكادميوم الثنائية . شخصت المركبات المحضرة بالطرق الطيفية المتوفرة (طيف الأشعة تحت الحمراء ، وطيف الأشعة فوق البنفسجية والمرئية) ، فضلا عن قياس العزم المغناطيسي المؤثر للمعقدات الصلبة . تم دراسة الاستقرارية للمعقدات المحضرة حيث وجد ان معقد الكادميوم هو الاكثر استقرارا" ومعقد الكروم الاقل استقرارا". درست الفعالية المضادة للجراثيم لكل من الليكاند ومعقداته على خمسة أنواع من البكتريا هي:
(Staphylococcus aureus, E. Coli, Proteus Vulgaris, Pseudomonas, and Klebsiella).

he genetic effects of several concentrations of L–Asparaginase II (ASNase II), produced by Proteus vulgaris strain Pv.U92, at various levels of purification (ultrasonication, precipitation, ion-exchange chromatography and gel filtration chromatography) on cancer cells line of Hep–2 were studied. This bacterial enzyme with concentration 4 U/ml at gel filtration level was revealed a putative cytotoxicity against cancer cells in comparison with other concentrations and steps of purification were used in this work. Moreover, 4 U/ml of ASNase II at gel purification level has a distinguished role on arrest cancer cells division of Hep–2; it was reduced the content of DNA at each phase of cancer cell cycle particularly at G2/M phase, the level of DNA was 3%. On the other hand, the partial purified enzyme, L–ASNase II, was induced apoptosis by both levels of purification ion–exchange and gel filtration, the apoptotic fractionation was 0.86 and 0.7 respectively .

درست التأثيرات الوراثية لتراكيز مختلفة من أنزيم L–Asparaginase II (ASNase II ) المنتج من السلالة البكترية Proteus vulgaris PVU92 وبمستويات مختلفة من التنقية ( الامواج فوق الصوتية ، الترسيب ، كروماتوكرافيا التبادل الايوني ، كروماتوغرافيا الترشيح بالهلام ) في الخلايا السرطانية من نوع Hep–2 . أظهر الستخلص البكتري بتركيز4 وحدة / مل وبمرحلة الترشيح الهلامي فعالية سمية في الخلايا السرطانية مقارنة مع بقية التراكيز ومراحل التنقية المستعملة في هذه الدراسة . فضلا عن ذلك كان للانزيم البكتري بنفس التركيز وبمرحلة الترشيح بالهلام دورا مميزا في أيقاف أنقسام الخلايا السرطانية Hep–2 حيث تمكن من خفض محتوى الدنا في كل طور من أطوارالأنقسام بحيث بلغ محتوى الدنا في طور G2/M 3% . ومن جانب اخر فقد تمكن الانزيم ASNase II المنقى جزئيا والمستعمل بمرحلتي التبادل الايوني و الترشيح بالهلام من تحفيز موت الخلايا السرطانية وقد بلغت النسبة المئوية لأجزاء الخلايا المنتحرة 0.86 , 0.7 على التوالي .

he aim of the study was to treat the pathogenesity and adhesion property of Proteus mirabilis isolate, obtained from urinary tract infection (UTI) patients. After the identification of Proteus mirabilis it was found that the pathogenic bacteria possessed the adhesion ability both in vivo and in vitro bioassays, with lactobacillus acidophilus filtrate as Probiotic. Proteus mirabilis was subjected to different concentrations of lactobacillus acidophilus filtrate to investigate its adhesion property and pathogenesity. Three concentrations (25, 50,100) % of Lactobacillus acidophilus concentrated filtrate were used in vitro bioassays against pathogenic bacteria. Results showed that the third fold 25% was the most effective concentration in reducing the adhesion intensity of the bacteria. This concentration was selected to be used in in vivo for detection of infectivity to the animal tissues (Kidney and liver) and also the effect of Proteus mirabilis and Probiotic on the level of hormones in liver; Serum glutamate pyruvate transaminase, Serum glutamate oxoloacelate transaminase and Alkaline phosphtases (GPT, GOT , ALP) and kidney enzyme (Urea) in serum of mice was investigated. It was found that the levels of liver hormones and kidney enzyme increased at the time of infection, and these levels returned to normal or decrease after treating the animals with 25% of concentrated filtrate of Probiotic.

يهدف البحث الى معالجة امراضية وصفة الالتصاق لبكتيريا المتقلبات والتي تم الحصول عليها من مرضى يعانون من التهاب المجاري البولية . بعد تشخيص البكتيريا الممرضة وجد بانها تمتلك قابلية الالتصاق وذلك من خلال الاختبارات الحيوية خارج وداخل الجسم الحي باستعمال الراشح الطبيعي المستخلص من بكتيريا اللبن , اخضعت البكتيريا الممرضة لثلاث تراكيز مختلفه من الراشح الطبيعي لبكتيريا اللبن وذلك لدراسة تاثير الراشح على صفة الالتصاق والامراضية . استعملت ثلاث تراكيز ( 25 ،50 ،100) % من الراشح الطبيعي المركز لبكتيريا اللبن في الاختبارات الحيويه خارج الجسم الحي . اوضحت النتائج بانه التركيز الثالث 25% كان التركيز الاكثر تاثيرا في خفض شدة التصاق البكتيريا الممرضة . انتخب هذا التركيز 25% في الاختبارات الحيوية داخل الجسم الحي وذلك لدراسة تاثيره على خفض امراضية البكتيريا الممرضة في الانسجة الحيوانية (الكبد والكلية) وعلى مستوى الهرمونات في الكبد وانزيم الكلية (اليوريا) في داخل جسم الحيوان( الفار) . اوضحت النتائج ارتفاع مستويات الهرمونات في الكبد واليوريا في الكلية عندما حقنت الفئران بالبكتيريا الممرضة وعادت الى مستوياتها الطبيعية او انخفضت قليلا عندما عولجت بالتركيز المثبط الثالث 25% لبكتيريا اللبن .

hronic myeloid leukemia (CML), also known as chronic granulocytic leukemia, is a form of leukemia characterized by the increased and unregulated growth of myeloid cells in the bone marrow and the accumulation of these cells in peripheral blood. Total RNA extraction, cDNA and quantitative real-time PCR (Q-RT-PCR) were done for thirty CML Iraqi patients. Hematology tests (hemoglobin, platelets and WBCs counts) were done for the same samples in the same time. The results of this study show that some samples with normal hematology values have BCR-ABL (p210) fusion transcript with molecular analysis by Q-RT-PCR. This indicates the importance of this technique in the diagnosis and monitoring the therapy of CML patients

سرطان الدم النخاعي المزمن احد أنواع السرطانات المنتشرة في العراق تتميز بالنمو غير المسيطر عليه لخلايا نخاع العظم وتراكم هذه الخلايا في الدم المحيطي . شملت هذه الدراسة ثلاثين عينة دم للمرضى تم استخلاص الرنا وتحويله الى cDNA ثم تم تضخيمه بجهاز Q-real time PCR . تم إجراء فحوصات الدم للمرضى وشملت اعداد كريات الدم البيضاء والهيموكلوبين وأعداد الصفائح الدموية . أظهرت نتائج الدراسة تعبير الجين قيد الدراسة BCR-ABL بنسب مختلفة لكنها لم تتناسب مع قيم فحوصات الدم

his study was conducted to investigate the efficiency of magnetic system in milk sterilization. This was done using bacteriological, (total microbial count) and cytogenetic (mitotic index and chromosomal aberration of bone marrow) investigations. Results indicated a reduction in total milk microbial count. On the other hand, mitotic index was increased and chromosomal aberration in bone marrow was decreased in magnetic milk as compared with raw milk. In conclusion, milk sterilization might performed using magnetic field application

أجريت هذه الدراسة للكشف عن كفاءة النظام المغناطيسي في تعقيم الحليب .واستخدمت الفحوصات البكتريولوجية (حساب العدد الكلي للاحياء المجهرية) وفحص الوراثة الخلوية (حساب معامل الانقسام والانحرافات الكروموسومية) في خلايا نخاع العظم للفئران .وقد اشارت النتائج الى انخفاض العدد الكلي للميكروبات ومن جانب اخر زيادة في معامل الانقسام مع انخفاض في عدد الانحرافات الكروموسومية في الحليب الممغنط مقارنة بالحليب الخام .نستنتج امكانية تعقيم الحليب باستخدام الحقول المغناطيسية .

his experiment was conducted in Science and Agriculture College labs, Kufa University, on 2008 to describe methods for extraction and purification of Procyanidin compound from each of grape seeds mother plant(Vitis vinifera L.) ,callus induced from Axillary buds by using MS medium supplemented with 2,4-D at concentration of (1,2,3)mg/l and from leaves of regenerated from callus by using MS medium supplemented with ( IAA )at concentration of 2mg/l and (BA) benzyl adenine at concentration of 0.2 mg/l and from cocoas powdered (control) as well these Procyanidin. The content of Procyanidin were done by using HPLC (high performance liquid chromatography) and compare of the quantitatively of Procyanidin with these seeds in mother plant and callus tissue. This study include alcohol extraction by using mixture of ethanol, methanol, D.W and HCL. The result revealed that, the superiority of Procyanidin in seeds content than the content in callus and regenerated plant and increase the content of these compound in the leaves of regenerated plant with the content in callus tissue.

أجريت هذه الدراسة في مختبرات كلية العلوم وكلية الزراعة / جامعة الكوفة عام 2008 وتضمنت استخلاص وتنقية مركب إلـ Procyanidin من مستخلص بذور العنب Vitis vinifera L ومن الكالس المستحث من البراعم الجانبية (Axillary's buds) باستخدام وسط MS المجهز بمنظم النمو2,4-D(Dichlorophenoxy acetic acid) تركيز 3,2,1 ملغم/لتر وكذلك من النموات المتولدة من أنسجة الكالس المستحث من البراعم الجانبية لنبات العنب باستخدام وسط MS المجهز بمنظمي النمو IAA( Indol acetic acid) تركيز 2 ملغم/لتر ول BA ( (Benzyl adenineتركيز 0.2 ملغم/لتر .جرى استخلاص كحولي للبذور والكالس والنموات المتولدة خارج الجسم الحي باستخدام مزيج من مذيبات الميثانول 80٪ وال الايثانول80٪ والماء المقطر وحامض .HCL تم التحري والكشف عن المركب الفينولي Procyanidin باستخدام كاشف خلات الرصاص 1٪.بينت النتائج بان المستخلصات الكحولية للعينات النباتية المدروسة قد أعطت كشفا ايجابيا مع خلات الرصاص وكانت نتائج التقدير الكمي لمركب Procyanidin قد بينت تفوق بذور العنب بمحتوى المركب تفوقا معنويا على محتوى انسجه الكالس مع تفوقا معنويا للمركب في المعاملة2,4-D تركيز 2ملغم/لتر مقارنة بمحتواه بالمعاملة 2,4-D تركيز1 , 3ملغم/لتر. نفذت التجربة باستخدام اختبار T (T– test) في مقارنة كمية المركب إلــ Procyanidin في الأجزاء النباتية المدروسة

hole blood samples were obtained from 30 pregnant women at 15 –24 weeks of gestation. DNA was extracted from each plasma or serum sample. To detect the Y-chromosome specific marker DYS14 in the maternal blood, (Polymerase Chain Reaction) PCR were carried out for each DNA extract. The PCR products were analyzed by 1.5% agarose gel electrophoresis and ethidium bromide staining. The results compared with fetal gender after delivery. The result of delivery revealed that 13 pregnant women had a male fetus and the remaining 17 pregnant women had a female fetus and DYS14 was detected in all plasma and serum samples obtained from pregnant women and revealed that 13 pregnant women had a male fetus and the remaining 17 pregnant women had a female fetus. The PCR sensitivity for detecting the gender of fetus from maternal whole blood at 15–24 weeks of gestation was 100% in both plasma and serum, DYS14 was not detected in the DNA from any of the 17 pregnant women carrying a female fetus. The results showed that PCR analysis of maternal plasma and serum can be used to diagnose fetal gender

جمعت 30عينة من الدم المحيطي لنساء حوامل (15-24) اسبوع من مدة الحمل , تم عزل DNA من كل من بلازما ومصل الدم , للكشف عن المؤشر الوراثي الخاص بالكروموسوم الذكريDYS14 باستخدام تقنية الـPCR , تم الكشف عن ناتج تفاعلPCR باستعمال الترحيل الكهربائي على هلام الاكاروز بتركيز 1,5 % , قورنت النتائج مع نتائج جنس الجنين ما بعد الوضع والتي اظهرت ان 13 مرأة كانت حامل بجنين ذكر و 17 المتبقية كانت حامل باناث , وذلك بالكشف عن المؤشر الوراثي DYS14 . اظهرت النتائج ان حساسية تفاعل الـ PCR في الكشف عن جنس الجنين في النساء الحوامل باستعمال الدم المحيطي للحوامل وبعمر حمل تراوح بين (15-24) اسبوع كانت 100% لكل من بلازما ومصل الدم . لم تظهر الواسمة DYS14 في اي من الـ 17 من النساء الحوامل بإناث . اظهرت النتائج امكانية استعمال PCR في الكشف عن جنس الجنين ما قبل الولادة

he aim of this study was to determine the effect of chemotherapy (vincristine and vinblastine) on the fertility and sex hormones level of female mice. Twenty four female albino mice were divided into three equal groups. The First group injected with vincristine 0.05 mg/kg intraperitoneally daily for 3 weeks. The second group injected with vinblastine 0.05 mg/kg intraperitoneally daily for 3 weeks. The third group treated with normal saline intraperitoneally daily for 3 weeks as control group. Blood samples were collected by using heart puncture within plastic syringes and transferred to plastic tubes. Blood centrifuged and serum was harvested for hormonal assay. The animals in each group were killed by dislocation of cervical vertebrate in third week of the treatment. Ovaries weighed and fixed with Bouin solution for histological study. Results showed significantly (P<0.05) decreased in ovaries weight on groups treated with vincristine and vinblastine as compared with control group. The mean numbers and diameters of primary, secondary follicles and corpus luteium, and hormones level [follicular stimulating hormone (FSH), lutenizing hormone (LH), estrogen and progesterone] were significant declined in groups treated with vincristine and vinblastine as compared with control group. Results indicated that fertility and sex hormones level reduced in females treated with chemotherapy drugs vincristine and vinblastine.

هدفت الدراسة الحالية الى معرفة تاثير بعض ادوية العلاج الكيمياوي (فنكرستين و فنبلاستين) على ، خصوبة اناث الفئران ومستوى الهرمونات الجنسية فيها . استخدمت في هذه الدراسة 24 انثى من الفئران قسمت الى 3 مجاميع متساوية ، حقنت المجموعة الاولى بالفنكرستين 0.05 ملغم/كغم داخل الخلب البريتوني يوميا لمدة ثلاثة اسابيع . اما المجموعة الثانية فقد حقنت بالفنبلاستين 0.05 ملغم/كغم داخل الخلب البريتوني يوميا لمدة ثلاثة اسابيع . في حين حقنت المجموعة الثالثة بالمحلول الملحي الفسيولوجي كمجموعة سيطرة داخل الخلب البريتوني يوميا لمدة ثلاثة اسابيع . تم جمع نماذج الدم عن طريق ثقب القلب بواسطة محاقن بلاستيكية ووضعت في انابيب بلاستيكية في جهاز الطرد المركزي للحصول على المصل لغرض قياس الهرمونات . تم قتل جميع الحيوانات عن طريق فصل الرقبة بعد الاسبوع الثالث من التجريع ، وزنت المبايض ووضعت في سائل باون لغرض اجراء الدراسة النسيجية . اظهرت النتائج انخفاض معنوي (P<0.05) في معدل اوزان المبايض في المجاميع المعاملة بالـفنكرستين والفنبلاستين مقارنة مع مجموعة السيطرة . وكذلك انخفضت معدلات اعداد واقطار الجريبات الاولية والثانوية والاجسام الصفراء ، ومستوى الهرمونات في الدم (الهرمون المحفز للجريبات والهرمون اللوتيني والاستروجين والبروجستيرون ) في المجاميع المعاملة بالـفنكرستين والفنبلاستين مقارنة

Abstract
A study aimed to examine immunomodulatory effect of concentrated cell free filtrate (CCF) of Lactobacillus acidophilus on immune system of mice by detecting the total and absolute counts of leucocytes. It was shown that CCF of Lactobacillus acidophilus caused a significant increase in total leucocyte count for all concentrations used in comparison with negative control. However, in absolute leucocyte counts the, CCF of L.acidophilus at doses (500 , 750) mg/kg caused a significant increased in lymphocyte, neutrophil and monocyte counts, while there was no significant differences in eosinophils and basophil cells number of mice treated with bacterial filtrate. The interaction study showed that the post-treatment with CCF of L. acidophilus caused a significant increase in total leucocytes count and absolute counts of all types of leucocyte cells except basophils and esinophils which showed no significant differences in comparison with pre-treatment.

هدفت الدراسة لمعرفة تأثير بكتريا حامض اللاكتيك التعايشية النوع Lactobacillus acidophilus على الجهاز المناعي للفئران من خلال تأثيرها على العد الكلي والتفريقي لكريات الدم البيضاء .بينت نتائج الدراسة إن المستخلص الخالي من الخلايا لبكتريا حامض اللاكتيك التعايشية النوع L. acidophilus سبب زيادة معنوية في العدد الكلي لكريات الدم البيضاء عند جميع التراكيز المستخدمة بالمقارنة مع السيطرة السالبة . وعند دراسة العدد التفريقي لكريات الدم البيضاء لوحظ إن بكتريا L. acidophilus سببت زيادة معنوية في خلايا الدم اللمفاوية والخلايا ألعدله وألأحاديه خصوصا عند التراكيز المرتفعة (500 و750) ملغم/كغم وزن جسم بينما لم تسبب زيادة معنوية في أعداد الخلايا الحامضية والقاعدية في الفئران المعاملة بالمستخلص البكتيري . أظهرت نتائج الدراسة التداخلية للمستخلص البكتيري مع العقار Cyclophosphomide أن المعاملة البعدية بالمستخلص البكتيري سببت زيادة معنوية في العد الكلي و التفريقي لكريات الدم البيضاء ما عدا الخلايا القاعدية والحمضية التي لم تتأثر بالمقارنة مع المعاملة القبلية بالمستخلص البكتيري .
