DIRECT DETECTION OF Clostridium perfringens BASED ALPHA TOXIN GENE FROM SHEEP AND CATTLE BY REAL TIME PCR TECHIQUE

Abstract

Real-time polymerase chain reaction assay was used for detection of
Clostridium perfringens toxins genes alpha (cpa )directly from the fecal samples of
cattle and sheep.Fecal samples from 20 clinically healthy cattle and 20 clinically
healthy sheep were collected randomly from different farms located in ALDiwanyia
city ,All the strains of Clostridium perfringens were analyzed by Real-
Time PCR using specific primers for alpha (cpa)toxin genes ,its reveald that
(cpa)toxin gene of Clostridium perfringens ,were detected in cattles in 8(40%) and
14 (70%) in sheep Real time positive respectively samples ,This results indicate that
Clostridium perfringens alpha toxin producing strains were prevalent in sheep and
cattles and its possibly play an important role in the diarrheal disease caused by
Clostridium perfringens .

استخدم اختبار االوقت الحقیقي لتفاعل سلسلة البلمرة للكشف عن جینات جرثومة المطثیات
مباشرة من عینات براز ل 20 من الابقار السلیمة سریریا (cpa) الحاطمة المسؤولة عن افراز سموم الفا
و 20 من الاغنام السلیمة سریریا جمعت عشوائیا من حقول مختلفة تقع في مدینة الدیوانیة .جمیع العزلات تم
تحلیلھا بواسطة اختبار الوقت الحقیقي لتفاعل سلسلة البلمرة باستخدام بادئات متخصصة لجینات السموم نوع
40 %) من ) لجرثومة المطثیات في 8 (cpa) تم من خلال الاختبار الكشف عن جینات سموم ،(cpa) الفا
70 %)من الاغنام عینات موجبة على `التوالي .النتائج تدل على ان عتر جرثومة المطثیات ) الابقار و 14
المنتجة لسموم الفا موجودة في الابقار والاغنام ومن المحتمل انھا تلعب دورا مھما في امراض الاسھال
الناجم عن جرثومة المطثیات الحاطمة .

Abstract

The Providencia rettgeri (P.rettgeri) is an opportunistic pathogen of human and
animals and has a clinical importance as a nosocomial pathogen causing infections in
the urinary system after catheterization and in immunocompromised patients less than
human feces, bile and sputum. To estimati the pathogenic role of P.rettgeri in mice
post-infection intraperitonial infection (I/P). thirty mice from both sexes, aged 6-7
weeks and weight 25-30 gm. were infected I/P by a whole bacterial suspension of
P.rettgeri 0.5 ml (containing 1x108 CFU/ml), the negative control group injected I/P
with (0.5 ml) PBS,five mice were sacrificed after (24, 48hrs. and 4,7 and 14 days)
post-inoculation, also (n=5) were considered as negative control group, for
histopathological examination. The histopathological changes at 24,48hrs until 4 days;
recorded mild to moderate acute inflammatory cells reaction, edema, fibrinous
exudate. At 7&14 days there was also necrosis and focal aggregation of mononuclear
cells. Strong pathogenic role of P.rettegri in induction acute inflammatory response in
experimental lab mice.

من الجراثیم الانتھازیة في الانسان والحیوان Providencia rettgeri (P.rettgeri) تعتبر بكتریا
على حد سواء لما لھا من اھمیة سریریة في قابلیتھا على احداث الخمج البكتیري الانتھازي في الجھاز البولي
عند استخدام القسطرة البولیة وفي حالات نقص المناعة المكتسب مع قلتھا في البراز والمرارة والقشع،لذاكان
في احداث التغیرات المرضیة النسیجیة P. rettgeri الھدف من الدراسةھو التعرف على الدور الخمجي لبكتریا
- 7 اسابیع واوزانھا 25 - في الفئران البیضاء المخمجة بھا داخل الخلب. تم استخدام ثلاثون فارا اعمارھا بین 6
30 غرام ومن كلا الجنسین قسمت عشوائیا الى خمسة مجامیع حسب فترات التضحیة بالحیوانات بعد تعریضھا
1خلیة بكتیریة/مل) داخل الخلب، و المجموعة السادسة تم حقنھا داخل x للخمج البكتیري بجرعة 0.5 مل ( 810
الخلب بالمحلول الملحي الفسلجي المعقم كمجموعة سیطرة سالبة. تم التضحیة بالحیوانات بعد انتھاء الفترات
الزمنیة للخمج البكتیري ( 24 و 48 ساعة وكذلك 4 و 7 و 14 یوما). اظھرت نتائج الخمج البكتیري خلال
48 ساعة وحتى 4 ایام ارتشاح الخلایا الالتھابیة الحادة في الاعضاء الداخلیة وتجمع السوائل ، الفترات 24
الخزبیة مع نضجة لیفینیة في الانسجة الخلالیة للاعضاء الداخلیة وغشاء الخلب، كما لوحظ نخر نسیجي مع
تجمعات الخلایا وحیدة النواةخلال الفترتین 7 و 14 یوما.نستتنتج مما سبق لضراوة الشدیدة لبكتریا
في احداث الطور الحاد من الاستجابة الالتھابیة في الفئران البیضاء والمصابة بھا داخل الخلب.

Abstract

This study was conducted in Basrah at period from September- 2012 throughout
August- 2013.Blood samples were collected from 480 sheep in different ages and
both genders. Haematological parameters were estimated and the result revealed that;
the sheep included in this study have lower haematological parameters than the
standard ones. Young animals in this study suffering from macrocytic normochromic
anaemia.

أجریت ھذه الدراسة في البصرة للفترة من أیلول 2012 إلى آب 2013 . جمعت نماذج الدم من 480 رأس
من الضأن من مختلف الاعمار ولكلا الجنسین وتم قیاس معاییر كریات الدم الحمر . أظھرت النتائج ان معاییر
كریات الدم الحمر في حیوانات الدراسة كانت اقل من المعیار العالمي القیاسي وان الحیوانات الصغیرة العمر
كانت تعاني من فقر الدم كبیر الخلیة وطبیعي الصباغ .
